Feasibility of an inhaled antibiotic/adjuvant dry powder combination using an experimental design approach

The global increase of multidrug resistant bacteria and the lack new classes antibiotic especially those targeting Gram-negative pathogens are leaving clinicians disarmed to treat numerous bacterial infections. Recently, design adjuvants able enhance antibiotics activities appears be one most promising investigated solutions circumvent this problem. In context, we have recently identified a polyamino-isoprenyl derivative NV716 potentiate, at very low concentration activity doxycycline against P. aeruginosa strains by increasing its intracellular concentration. study will report an experimental protocol optimize dry powder for inhalation ensuring simultaneous delivery (doxycycline) adjuvant (the polyaminoisoprenyl since aerosol therapy could allow rapid drug administration target respiratory system avoiding first pass effect minimizing undesirable systemic effects. Thus, was carried out permitting identify influence several factors on aerosolization efficiency our combination allowing us find right composition manufacture leading best optimization two compounds in form inhalable powder. More precisely, powders active ingredients were prepared freeze drying their improved addition carrier particles lactose grade. Under these conditions, formulation defined combining optimal aerodynamic properties’ values lowest MMAD (Mass Median Aerodynamic Diameter) highest FPF (Fraction Fine Particles)) without even using sophisticated engineering techniques. Finally, results suggest that molecules successfully delivered requested lungs then decrease consumption as well treatment efficacy.
